The actors, both 29, will create new comedy characters and perform as themselves in front of a live studio audience in the BBC3 series.
The show, which has been given the working title Horne and Corden Have Come, will be directed by Kathy Burke and written by Corden and Horne themselves.
For Burke, the actress-turned theatre-director best known for her role as Waynetta Slob in the Harry Enfield TV series, it will be her first TV project behind the camera.
News of the commission comes after Gavin and Stacey, created by Corden and Ruth Jones, won the audience award for best programme at the weekend's TV Baftas.
