Edited by Stuart (biological sciences, Brock U., Canada), this is the second edition of a work offering established protocols and newer methods for studying aspects of mitochondirial DNA. The first group of papers discusses protocols used to study the transduction of information from mitochondrial DNA to functionally active respiratory complexes, covering investigations of the nucleoid proteome, mitochondrial DNA packaging, replication, transcription, respiratory complex synthesis, and polymerase gamma mutations associated with mitochondrial disorders. The next section focuses on mitochondrial reactive oxygen species production, mitochondrial DNA damage, and its repair. The final section examines new approaches for identifying and quantifying heteroplasmic mitochondrial DNA mutations, which may be associated with some of the bioenergetic deficits observed in age-associated disease. As with other volumes in the series, each paper includes introductions to the topic, lists of necessary materials and reagents, step-by-step protocols, and notes on troubleshooting and pitfalls.
