This edition of a lab manual for a yeast genetics course retains its exclusive focus on the baker's yeast, S. cerevisiae. For this edition, the gene replacement experiment has been modernized to include new drug-resistance markers. In the Techniques and Protocols sections, a tag protein purification protocol has been added, and simpler and more reliable protocols for extracting proteins from yeast and for colony PCR are included. There is a new section on handling large collections of yeast strains. Amberg is affiliated with the State University of New York's Upstate Medical University.
