Antibody detection with Blastomyces dermatitidis lysate antigens prepared from dog isolates.

Blastomycosis, a potentially lethal fungal disease of humans and animals, is caused by the thermally dimorphic organism Blastomyces dermatitidis. Diagnosis of this disease has been a challenge to clinicians for many years. The aim of this study was to evaluate B. dermatitidis yeast lysate antigens, prepared from three dog isolates (T-58 Tennessee, ERC-2 Wisconsin, WI-R Wisconsin), when the yeast cells were allowed to lyse in distilled water for one or seven days. The indirect enzyme-linked immunosorbent assay (ELISA) was used to determine the ability of the lysate antigens to detect antibodies in dog (18) and rabbit (18) serum specimens. When reacted with dog and rabbit sera, day 7 lysates produced greater reactivity than that of the day 1 lysates. The absorbance values ranged from 0.524 (T-58) to 0.651 (WI-R) with day one preparations when assayed against the rabbit sera and the day seven antigens ranged from 0.660 (WI-R) to 0.863 (ERC-2). When the dog sera were assayed with the day one lysates, the absorbance values ranged from 0.375 (ERC-2) to 0.547 (T-58) whereas day seven lysates exhibited absorbance values of 0.464 (WI-R) to 0.776 (T-58). Therefore, these dog lysate antigens exhibit the potential of being a useful immunodiagnostic tool for the detection of blastomycosis.
