Simplify and Accelerate Cloning.


Promega Introduces Thermosensitive Alkaline Phosphatase alkaline phosphatase /al·ka·line phos·pha·tase/ (ALP) (fos´fah-tas) an enzyme that catalyzes the cleavage of orthophosphate from orthophosphoric monoesters under alkaline conditions.  

MADISON, Wis. -- Scientists can now save time and simplify the cloning process with the new Promega Thermosensitive Alkaline Phosphatase (TSAP TSAP - Transport Service Access Point ). For dephosphorylation of no DNA DNA: see nucleic acid.

One of two types of nucleic acid (the other is RNA); a complex organic compound found in all living cells and many viruses. It is the chemical substance of genes.
 in cloning reaction, scientists get both speed and simplicity because two unique features found in TSAP:

1. TSAP is active in all restriction enzyme restriction enzyme

Protein (more specifically, an endonuclease) produced by bacteria that cleaves DNA at specific sites along its length. Thousands have been found, from many different bacteria; each recognizes a specific nucleotide sequence.

As a quality product, TSAP is blue/white cloning-qualified. Scientists get a higher level of quality control for enzymes used in cloning applications. TSAP catalyzes the removal of 5' phosphate groups from DNA, thus preventing the recircularization and religation of linearized cloning vector DNA during ligation. It is effective on 3' and 5' overhangs and on blunt ends.
