Peptide Composer is a composition-based sequencing (CBS) software package available to all FT-ICR mass spectrometer users. CBS is a method for the mass spectrometrical determination of the amino acid sequence of unknown peptides, which are not necessarily listed in protein (or other) databases. With the availability of ultraprecise mass spectrometrical biomolecular data, the accurate mass is said to be becoming a physical quantity of high interest in bioanalytical methodology. FT-ICR mass spectrometry with ESI or MALDI sources is said to now allow one to easily determine the amino acid composition of medium sized unknown peptides when employing combinatorial calculation of hypothetical parent and fragment ion masses. This new method, which in a second step allows one to reliably sequence completely unknown peptides, appears to open a wide new field of bioanalytical investigation. It is said to be clear that, while the nominal mass of a peptide has obviously no useful correlation to biomolecular structure, the accurate mass indeed has a strong and direct correlation that has not been exploited considerably so far. Strategies for extracting structural and even functional information out of accurate mass values of biomolecules have to be developed in the future, according to the company. The CBS approach is said to be highly efficient and very fast.
