Recent Independent Study Shows Microfluidizer Processors Speed Development of Gene Therapies to Produce Recombinant Adenoviruses.

Business/Biotechnology Editors & Health/Medical Writers

NEWTON, Mass.--(BW HealthWire)--April 16, 2001

Adenoviruses are genetically modified viruses used in gene therapy to treat cystic fibrosis cystic fibrosis (sĭs`tĭk fībrō`sĭs), inherited disorder of the exocrine glands (see gland), affecting children and young people; median survival is 25 years in females and 30 years in males. , cancer and (potentially) other diseases. One of the more time consuming steps in the manufacture of recombinant Adenoviruses (rAd) is the manual lysing (disruption) of mammalian cells in which rAd are cultivated.

Freeze-thaw cycles are commonly used to lyse lyse (liz)
1. to cause or produce disintegration of a compound, substance, or cell.

2. to undergo lysis.

mammalian cells, taking up to 6 hours to process a 1.5-liter batch of cellular material. However, in a recent comparative study by Cobra Therapeutics in Staffordshire, England, the Microfluidizer(R) materials processor, processed the same size batch in only 2 hours, with no detrimental effects on the virus yields or infectivity. The continuous processing capability of the Microfluidizer processor provides additional safety benefits as it limits human interaction with the cells, reducing the risk of employee contamination. Processing larger amounts of cell suspensions results in even greater savings of time and labor expenses.

The Microfluidizer processor is based on a patented high-shear fluid processing technology, used for cell lysing and the preparation of vaccines, parenterals, encapsulation (1) In object technology, the creation of self-contained modules that contain both the data and the processing. Microfluidizer processors are available in lab, pilot and production scale units and are currently used by most leading biotech, genomics, pharmaceutical and biopharmaceutical companies, worldwide, in both cell lysing and drug formulation applications.
