Effects of estradiol 17-beta and tamoxifen on MCF-7 human breast cancer cells.

 are considered to be probable female carcinogens and have impact on reproductive system. We have examined in vitro effects of estradiol 17-beta, and tamoxifen on estrogen responsive MCF-7 human breast cancer cells. Estradiol 17-beta ([10.sup.-]6 and [10.sup.-4] M) and tamoxifen ([10.sup.-6] and [10.sup.-4] M) were added to the cultures of MCF-7 cells, and incubated for 24 hours Adv. 1. for 24 hours - without stopping; "she worked around the clock"

. Morphological changes were observed under phase-contrast microscope. The MTT assay was performed to assess cell viability. We have observed very low level of cell viability with [10.sup.-4] M tamoxifen when compared to the DMSO DMSO dimethyl sulfoxide.

 control (P <0.05). There was a minor increase in cell viability with [10.sup.-6] M estradiol 17-beta. However, [10.sup.-4] M tamoxifen again significantly inhibited (P <0.01) the cell viability when compared to [10.sup.-6] M estradiol 17-beta. These results indicate that exposure of estrogen and tamoxifen to human breast cancer cells yielded changes in cell shape and cell viability. This may provide a useful model to study steroid hormone regulation of cytoskeleton cytoskeleton
