Daiichi Sankyo Files New Drug Application for Amoldipine Beslyte, Olmesartan Medoxomil Combination for Hypertension Treatment.


Tokyo, Japan, Nov 30, 2006 - (JCN) - Daiichi Sankyo announced November 29 that it has filed a New Drug Application (NDA) with the U.S. Food and Drug Administration (FDA) for a fixed dose combination of two antihypertensives antihypertensive /an·ti·hy·per·ten·sive/ (-ten´siv) counteracting high blood pressure, or an agent that does this.

) amlodipine amlodipine /am·lo·di·pine/ (am-lo´di-pen?) a calcium channel blocking agent used as the besylate besylate /bes·y·late/ (bes´i-lat) USAN contraction for benzenesulfonate. salt in the treatment of hypertension and chronic stable and vasospastic angina. besylate (currently marketed by Pfizer as Norvasc) and the angiotensin angiotensin /an·gio·ten·sin/ (-ten´sin) a decapeptide hormone (a. I) formed from the plasma glycoprotein angiotensinogen by renin secreted by the juxtaglomerular apparatus. It is in turn hydrolyzed by a peptidase in the lungs to form an octapeptide (a. II), which is a powerful vasopressor and stimulator of aldosterone secretion by the adrenal cortex. This is in turn hydrolyzed to form a heptapeptide (a. receptor blocker (ARB) olmesartan medoxomil, or Benicar.

Pending approval, the combination will be indicated for the treatment of hypertension and may be used either alone or in combination with other antihypertensive agents.

In a clinical trial, the amlodipine/olmesartan combination demonstrated clinically significant mean blood pressure reductions. It utilizes two complementary mechanisms of action, calcium channel blockade and angiotensin II angiotensin II
